Dr Joshua Boateng obtained his Bachelor of Pharmacy degree from the Kwame Nkrumah University of Science and Technology in February 1999 (First Class Honours) and then pursued an MSc in pharmaceutical analysis programme at the University of Strathclyde under a DFID Commonwealth Scholarship, graduating in November 2000. He subsequently undertook PhD research in pharmaceutical sciences (drug delivery and formulation) under a Pfizer-sponsored project at the University of Strathclyde, graduating from 2001-05. Subsequently, he took up a postdoctoral position at the Nottingham Trent University as part of a WADA-sponsored consortium on investigating surrogate biomarkers for detecting gene doping in sports, using proteomic approaches (2005-07).

Dr Boateng still maintains active research interests in all the above areas and published extensively as well as presented at several conferences around the above themes. He joined the School of Science at the University of Greenwich in August 2007 and is currently a principal lecturer in formulation science and formulation technology.

Dr Boateng belongs to the pharmaceutical science research cluster within the School of Science with researchers spanning pharmaceutical, analytical and material sciences. He works in an interdisciplinary range of research areas covering pharmaceutics (formulation and drug delivery), pharmaceutical analysis and proteomics.

Development of Novel Formulations for Buccal Mucosa Delivery of Protein Based Drugs

Background and justification

Protein based drugs offer great potential for better and effective therapeutics, but limited by rapid clearance from the body, immunogenicity and denaturation. They do not easily cross biological membranes making bioavailability low. Currently, most protein based drugs are usually administered by injection, which is inconvenient and leads to non-compliance. Difficulties encountered in the administration of protein-based drugs provide the motivation for developing novel formulations capable of maintaining therapeutic levels without side effects as well as traversing the deleterious gastro-enteric environment.

The need for protein based therapy such as vaccines is intense as mortality and morbidity are increasing for most infectious diseases especially in Africa. For example, around two to three million lives are lost yearly due to malaria, with approximately 90% of these in Sub-Saharan Africa. The implications of an increasing burden on the public health infrastructure and economic stability of Ghana is cause for concern. As a result, alternatives to the currently available treatment options that focus on prevention strategies such as better delivering vaccines have become research priorities. There is therefore the need to develop novel formulations for mucosal delivery of protein based drugs to reduce or eliminate the problems with the currently available pharmaceutical therapies.

Key findings

Optimised xerogels have been developed by freeze-drying chitosan based gels loaded with model protein and peptide drugs and functionally characterised. In vitro and ex vivo studies showed their ability to maintain the stability of the protein based drugs during and after formulation, release of the protein and subsequent permeation through tissue engineered buccal mucosa cells and sheep buccal mucosa.

Plans are in place to undertake in vivo studies in animals and to apply in relevant areas such as mucosal vaccination including paediatric candidates.
